Is it possible to somehow use the same setup on multiple computers?
If yes, is it possible to have the parameter file synced such that changes made to one computer TS2 setup are updated on the other one?

You could copy you config file ~/Library/Preferences/com.binaryage.TotalSpaces2.plist between computers.
This would not work for things directly connected to the desktops though (such as app-desktop assignments) because they are stored by OSX, and they are stored against the exact desktop UUID (so will be different on different machines).
I’m sorry, that’s probably not much help.
It would be possible to extract and set certain settings such as desktop names using the API https://github.com/binaryage/totalspaces2-api/tree/master/ruby , but the app assignments are not included at the moment.

Can you explain a bit more what you mean?
Go into System Preferences > Mission Control, and uncheck “Show Dashboard as a Space”. Should no longer show up in your TS grid view.
